Magnetic bead-based assays are a trusted method for multiplex analysis. However, traditional readouts using flow-cytometry-based platforms come with a range of frustrations: fluidics management, daily maintenance, calibration, and the ever-present risk of clogs causing bead failures. With Mabtech EYRA™, we reimagined this process by replacing fluidics with confocal microscopy.

Once your EYRAplex assay is finished and the beads have settled on the bottom of the wells, you simply insert the 96-well plate into Mabtech EYRA™ and press “read.” There’s no warm-up time, no priming, and no risk of sample loss - the sample remains in the same well during data acquisition.

During data acquisition, magnetic bead complexes remain undisturbed at the bottom of each well. EYRA’s internal lasers excite each bead’s unique dye signature, revealing its fluorescent Bead ID. Simultaneously, excitation of the PE detection signal reflects the amount of analyte bound to the bead.

What happens next is where the innovation shines. These emissions are captured as high-resolution images by the integrated confocal microscope. From here, Opal™ software takes over. Building on our experience with the RAWspot™ algorithm used in Mabtech ASTOR™ and IRIS™, we developed RAWsphere™. Using this algorithm, Opal™ automatically identifies each bead, correctly determines its bead-ID, and matches it with the corresponding PE-signal. The result is highly accurate multiplex data acquisition without the stress, downtime, or overhead of flow-based systems.

Finally, the readout delivers what you care about most: quantitative, publication-ready data. Standard curves are generated in Opal™, and the PE-signal is translated into analyte concentrations. The software instantly delivers these values into a single Excel output complete with standard curves, sample MFI values, and concentrations, making it easy to interpret complex immune responses at a glance.
