This peptide pool is intended for studying immune responses against adenovirus serotype 5 (AdV5) using immunoassays such as ELISpot and FluoroSpot. It induces cytokine secretion from antigen-specific human T cells. For research use only. Not for use in diagnostic procedures.

Description
This AdV5 (hexon) peptide pool induces secretion of e.g., IFN-γ and IL-2 from human T cells. It contains 94 peptides derived from the human adenovirus 5 (hAdV5) hexon protein. It consists of both defined and scanning peptides. The scanning peptides are 15-mers overlapping with 11 amino acids. Together, these peptides cover immunodominant regions of the hAdV5 hexon protein that share sequence homology to human adenovirus 26 and chimpanzee adenovirus Y25. The mean purity of the synthetic peptides is 80%.
The peptide pool has been validated in ELISpot and FluoroSpot using human PBMC.

Performance
Human PBMC (250,000 cells/well) were incubated overnight in the absence or presence of the Adenovirus 5 hexon peptide (Mabtech) and the number of cells secreting IFN-γ was analyzed by ELISpot.
Human PBMC (250,000 cells/well) were incubated overnight in the absence or presence of the Adenovirus 5 hexon peptide pool (Mabtech) and the number of cells secreting IFN-γ and/or IL-2 was analyzed by FluoroSpot. Single cytokine secreting cells as well as double-secreting cells were determined. Spot center was used to identify spots from double-secreting cells, visualized in an image overlay of LED490, FITC (IFN-γ) and LED550, Cy3 (IL-2). Spot analysis was made with Mabtech IRIS.
